• Ordinarily F-2, 6-BP binds to and increases the activity of phosphofructokinase 1.
• For example, during glycolysis, fructose-6-phosphate is converted to fructose-1, 6-bisphosphate in a reaction catalysed by the enzyme phosphofructokinase 1 ( PFK-1 ).
• Phosphofructokinase mutation and cancer : In order for cancer cells to meet their energy requirements due to their rapid cell growth and division, they survive more effectively when they have a hyperactive phosphofructokinase 1 enzyme.
• When exposed to aerobic conditions, the ATP and Citrate production increases and the rate of glycolysis slows, because the ATP and citrate produced act as allosteric inhibitors for phosphofructokinase 1, the third enzyme in the glycolysis pathway.
